Benedict’s test is a biochemical method used to detect the presence of reducing sugars (such as glucose, lactose, maltose, etc.) in a sample. It works by reducing Copper(II) ions in the reagent to a red or orange Copper(I) oxide precipitate under alkaline and heated conditions.

Preparation of Benedict's Reagent

To prepare 1 litre of Benedict’s reagent, we need the following components:

Procedure: Dissolve sodium citrate and sodium carbonate in approximately 800 mL of distilled water using a hot plate or stirrer. Then, in a separate beaker, dissolve copper sulfate in 100mL distilled water. Now, slowly add copper sulfate solution to carbonate-citrate solution while stirring constantly. Finally, allow the mixture to cool and make up the volume to 1 Litre with distilled water.

Preparation of Test Tubes

  1. Wash the test tubes properly and rinse it with distilled water.
  2. Label the test tubes.
  3. Test tube A: Sample solution
  4. Test tube B : Control(distilled water)

Mixing Ratio:

Now, mix gently by shaking carefully. At this stage the solution should be blue.

Control: Always prepare a negative control (distilled water + reagent) to verify reactivity. Test tube B is the control.

Inubation (Heating)

Place the test tube A in a boiling water bath at approximately 85 to 100 °C and heat for 3 to 5 minutes. Remove the test tube carefully by using a test tube holder.


Allow it to cool slightly to observe the color change and precipitate formation clearly.



Now observe the color changes and if you observe the following colors then there inference will be:

If the solution changes from blue to any of the above mentioned colors(except remaining blue) it means reducing sugars are present in it while if the color remains blue it indicates that reducing sugars are not present(absent).
